چکیده

We found the alpha-particle priming dose which could effectively induce in vivo radioadaptive response in the embryos of the zebrafish, Danio rerio, to be 4.8e48 mGy, through the apoptotic signals scored in the embryos at 24 h post fertilization. Specially fabricated thin polyallyldiglycol carbonate (PADC) films were used as support substrates for the dechorionated embryos. During irradiation, alpha particles passed through the PADC films to hit the zebrafish embryos, and the alpha-particle tracks were revealed in the films through chemical etching. By determining the number of alpha-particle tracks that coincided with the effective areas of the embryos, the alpha-particle doses were found. 2011 Elsevier Ltd. All rights reserved.

Alpha-particle-induced bystander effects between zebrafish embryos in vivo

Dechorionaed embryos of the zebrafish, Danio rerio, at 1.5 h post-fertilization (hpf) were irradiated with alpha particles from an 241Am source. Thin polyallyldiglycol carbonate (PADC) films with a thickness of 16 mm were used as support substrates for holding the embryos and recorded alpha-particle hit positions, and thus enabled calculation of the dose absorbed by the embryos. The irradiated ...

Radioadaptive response induced by alpha-particle-induced stress communicated in vivo between zebrafish embryos.

We report data demonstrating that zebrafish embryos irradiated by alpha particles can release a stress signal into the water, which can be communicated to the unirradiated zebrafish embryos sharing the same water medium and thereby inducing a radioadaptive response in these unirradiated zebrafish embryos. The effects of radiation on the whole embryos were studied through quantification of apopt...

Designing experimental setup and procedures for studying alpha-particle-induced adaptive response in zebrafish embryos in vivo

The present work was devoted to designing the experimental setup and the associated procedures for alpha-particle-induced adaptive response in zebrafish embryos in vivo. Thin PADC films with a thickness of 16 lm were fabricated and employed as support substrates for holding dechorionated zebrafish embryos for alpha-particle irradiation from the bottom through the films. Embryos were collected w...

Fabrication of pseudo three-dimensional PADC cell culture substrates for dosimetric studies

Pseudo three-dimensional (3D) polyallyldiglycol carbonate (PADC) cell culture substrates were fabricated by creating micrometer-size pores on thin PADC films by alpha-particle irradiation and subsequent chemical etching. Our results showed comparatively fewer 53BP1 foci in alpha-particle irradiated as well as bystander HeLa cells cultured on pseudo 3D PADC substrates than in those cultured on t...

Non-induction of radioadaptive response in zebrafish embryos by neutrons

In vivo neutron-induced radioadaptive response (RAR) was studied using zebrafish (Danio rerio) embryos. The Neutron exposure Accelerator System for Biological Effect Experiments (NASBEE) facility at the National Institute of Radiological Sciences (NIRS), Japan, was employed to provide 2-MeV neutrons. Neutron doses of 0.6, 1, 25, 50 and 100 mGy were chosen as priming doses. An X-ray dose of 2 Gy...
